Using AVG Free Edition...

Start the application using the desktop icon (if you chose to have it placed there) or from Start | Programs | AVG 6.0 Anti-Virus System (the default location, if you changed it, open it from there)

Grisoft's AVG Free Edition startup screen
The startup screen! Pretty!

AVG - run complete test
Click on the 'Run Complete Test' button to start the virus scan

AVG - Scanning Screen
The scanning screen. Gives some progress indication as well as notification of number of viruses and name of the last virus found.

Once the scan is complete, you will be given the option of what to do with infected files - delete them or quarantine them in the virus vault. AVG will tell you what it recommends doing, but leaves the choice up to you. In the event of serious infection, you may need to run AVG from the command line after a restart. Check the help documentation from within AVG, or go to www.grisoft.com for even more information and help.

In case you are wondering if a FREE application can compete with the 'big boys', see this review by 'Maximum PC'.

I have been using this software and recommending it for several years. You can't beat the price and it does a great job to boot!
